predvsem v zimskem obdobju. Najpomembnej\u0161i biotski dejavniki, ki vplivajo na odmiranje dru\u017ein je zunanji zajedalec Varroa destructor in \u010debelji virusi, ki jih ti zajedalci prena\u0161ajo. Zdravljenje virusnih obolenj, ki jih prena\u0161ajo varoje je ve\u010dinoma posredno, s prepre\u010devanjem razvoja varoj in njihovim zatiranjem. V dosedanjih raziskavah smo uspeli z razvojem selekcije \u010debeljih dru\u017ein na osnovi odpornosti proti virusom. Poznavanje in odkrivanje lastnosti zalege trotov, ki je prosta virusnih oku\u017eb, omogo\u010da ugotavljanje in odbiro odpornih dru\u017ein. Tako imenovana lastnost &#8216;zmanj\u0161evanja virusne infekcije jaj\u010dec&#8217; (SOV) ka\u017ee zmerno stopnjo dednosti za pojavljanje virusa deformiranih kril pri \u010debelah (h2 = 0.22). Predlog projekta predstavlja nadaljevanje uporabe SOV v \u010debeljih dru\u017einah, z ugotavljanjem genetske predispozicije za to izra\u017eeno lastnost. V ta namen bomo kri\u017eali odporne in dovzetne linije \u010debel, in testirali genetski material (haplotipe) v drugi generaciji zalege trotov (ko se star\u0161evski aleli razdelijo v genomu haploidnih potomcev), ter sekvencirali eksone celotnega genoma in razvoj genetskih lastnosti. Nadalje bomo prou\u010devali mo\u017enost oblikovanja odpornih linij \u010debel z uporabo naslednje generacije spreminjanja genov (CRISPR\/Cas9). Na osnovi izvedenih poskusov bomo izvedli raziskavo med \u010debelarji in vzrejevalci matic o mo\u017enosti uporabe nove tehnologije v vzrejni dejavnosti v \u010debelarstvu. V kon\u010dni fazi bomo preskusili ali se za\u017eeljene genetske lastnosti na osnovi SOV obdr\u017eijo v populaciji \u010debel in v razli\u010dnih okoljih.<\/p><p class=\"MsoNormal\" style=\"margin-bottom: .0001pt; Pridobili smo ustrezno \u0161tevilo vzorcev, ki smo jih preiskali na prisotnost \u010debeljih virusov. Pridobljeni rezultati vmesnega raziskovalnega obdobja so pomembni za potrditev pojavljanja vertikalnega in horizontalnega prenosa virusov pri medonosni \u010debeli.<\/li>\n<li>ugotavljanje napadenosti \u010debeljih dru\u017ein z varojami in zatiranje varoj v dru\u017einah.<\/li>\n<li>prou\u010devanje virusnih oku\u017eb v razvijajo\u010dih se \u010debeljih maticah. Za ta namen smo izvedli poskuse in vzor\u010denje jaj\u010dec, li\u010dink, bub in neopra\u0161enih matic. Detekcija virusov v zalegi je v teku<\/li>\n<li>vzor\u010denje zalege, jaj\u010dec trotov ki so haploidni. Najpomembnej\u0161i biotski dejavniki, ki vplivajo na odmiranje dru\u017ein je zunanji zajedalec Varroa destructor in \u010debelji virusi, ki jih ti zajedalci prena\u0161ajo. Zdravljenje virusnih obolenj, ki jih prena\u0161ajo varoje je ve\u010dinoma posredno, s prepre\u010devanjem razvoja varoj in njihovim zatiranjem. V dosedanjih raziskavah smo uspeli z razvojem selekcije \u010debeljih dru\u017ein na osnovi odpornosti proti virusom. Poznavanje in odkrivanje lastnosti zalege trotov, ki je prosta virusnih oku\u017eb, omogo\u010da ugotavljanje in odbiro odpornih dru\u017ein. Tako imenovana lastnost &#8216;zmanj\u0161evanja virusne infekcije jaj\u010dec&#8217; (SOV) ka\u017ee zmerno stopnjo dednosti za pojavljanje virusa deformiranih kril pri \u010debelah (h2 = 0.22). Predlog projekta predstavlja nadaljevanje uporabe SOV v \u010debeljih dru\u017einah, z ugotavljanjem genetske predispozicije za to izra\u017eeno lastnost. V ta namen bomo kri\u017eali odporne in dovzetne linije \u010debel, in testirali genetski material (haplotipe) v drugi generaciji zalege trotov (ko se star\u0161evski aleli razdelijo v genomu haploidnih potomcev), ter sekvencirali eksone celotnega genoma in razvoj genetskih lastnosti. Nadalje bomo prou\u010devali mo\u017enost oblikovanja odpornih linij \u010debel z uporabo naslednje generacije spreminjanja genov (CRISPR\/Cas9). Na osnovi izvedenih poskusov bomo izvedli raziskavo med \u010debelarji in vzrejevalci matic o mo\u017enosti uporabe nove tehnologije v vzrejni dejavnosti v \u010debelarstvu. V kon\u010dni fazi bomo preskusili ali se za\u017eeljene genetske lastnosti na osnovi SOV obdr\u017eijo v populaciji \u010debel in v razli\u010dnih okoljih.<\/p>\n<p><strong>\u00a0<\/strong><\/p>\n<p><strong>Abstract EN<\/strong><\/p>\n<p>Honey bees are under pressure due to abnormal high death rates, especially during the winter. The infestation by the Varroa destructor mite and the viruses that this ectoparasite transmits are generally seen as the most important biotic causes. The control of these mite-transmitted viruses is almost exclusively done in an indirect way, by limiting the severity of the mite infestation. We recently succeeded in the selection towards virus resistant honey bee colonies, based on a novel trait that renders honey bee drone brood eggs free of virus infections. This so-called \u2018suppressed in ovo virus infection\u2019 (SOV) showed moderate heritabilities for the load of Deformed wing virus (h2 = 0.22). The present project will further elaborate this SOV trait by mapping its genetic predisposition in bees. Therefore we will cross resistant and sensitive strains, test the phenotype in the F2 drone brood (when the parental alleles are segregated in the haploid genome), and perform whole exome sequencing and elastic regression. Further, we will test whether resistant bees can also be created by gene-editing using the CRISPR\/Cas9 technology. In concert with this, we will start a social study to determine the extent to which there is social support to implement this technology in the honey bee&#8217;s breeding programs.
